【摘要】:ThisguideisintendedforuseinabiotechnologylaboratorywheneverthenecessityarisesforidentifyingabiologicalpreparationbelievedtocontainprimarilyHSVoritsDNA.Thecharacterizationcriteriausedfortheidentificationshallbeperformedbyanindividualtrainedinmolecularvirology.ThisguideisnotmeanttobeusedinaclinicallaboratoryfortheidentificationofHSVisolatedfrompatientspecimens.1.1ThisguidecoverslaboratorycharacterizationproceduressufficienttoidentifypurifiedspecimensofHSVtypes1and2(HSV-1andHSV-2)orHSV-1DNAandHSV-2DNAusedinbiotechnology.ForcasesinwhichidentificationofHSVDNAspecimensisrequired,thecharacterizationcriteriaofandofthisguidearesufficient.1.2ThisguidedoesnotcovertheidentificationofHSVinHSV-infectedhostcells.Toapplythisguidetosuchacase,itwouldfirstbenecessarytoisolatethevirusfromsuchsamplesusingstandardtechniquesofHSVpurification.ThisguidedoesnotcovercharacterizationofsegmentsofHSVDNAorofvectorscontainingHSVDNAsegments.1.3Thisguidedoesnotcoverthespecificmethodologyusedintheidentificationcharacterization.Itdoesnotaddressthequestionofdegreeofpurityrequiredforherpesviruspreparations:thiswouldvarydependingontheparticularbiotechnologyuseofthevirus.1.4Warning-Laboratoryworkinvolvingherpessimplexvirusescanbehazardoustopersonnel.Biosafety2levelfacilitiesarerecommended().SafetyguidelinesshallbeadheredtoaccordingtoNCCLSM29-T2andotherrecommendations().Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
